Loop Mediated Isothermal Amplification (LAMP)

1. LAMP amplifies DNA under isothermal conditions.
2. LAMP amplifies DNA with high specificity by recognizing six sites on the target sequences.
3. LAMP amplifies DNA with such high efficiency that it enables detection of only a few copies of DNA in a short time. Actually, with incubation at 60℃ for 45min, only six copies HBV DNA was able to be detected by agarose gel electrophoresis followed by staining with SYBR Green I.
4. LAMP amplifies unique structural products of inverted repeat of DNA.
5. LAMP amplifies target RNA at an isothermal temperature using thermo-stable reverse transcriptase (RT-LAMP). Thus, it was capable of detecting only a cell among 106 cells, expressing mRNA of Prostate Specific Antigen.
Furthermore, we have developed simple and easy detection methods based on the characteristic features of LAMP. In conclusion, the results described above suggest that LAMP serves as a very useful method for DNA diagnostics.
