A Novel SNP Typing Technology Based on the Nucleic Acid Amplification Method, LAMP.

A variety of SNP typing technologies have been so far reported. However, more convenient and accurate methods are now required. Loop-mediated isothermal amplification (LAMP) is a novel nucleic acid amplification method that amplifies DNA specifically and rapidly under isothermal conditions (NAR, 2000, Vol.28, No.12, e63).
Recently we have found that specially designed primers of LAMP discriminated a single nucleotide change at each cycling step of the DNA replication. Thus, type of SNP can be easily detected by the experiment to find if the DNA containing the SNP has amplified. Actually, CYP2C19*1 and CYP2C19*2 alleles of drug responsive P450 gene were easily and strictly distinguished each other and were also discriminated from their homologus genes, CYP2C9 or CYP2C18.

The LAMP based SNP typing is an accurate, easy and inexpensive method to be operated. The usefulness of SNP typing technology, which is not dependent on PCR, will be discussed at the session.
