Visual detection

Visual detection by turbidity

The turbidity of magnesium pyrophosphate, a by-product of the amplification reaction, is produced in proportion to the amount of amplified products. Since LAMP amplification can produce extremely large amount of amplified products, white turbidity can be visually observed. From this feature, the presence of turbidity can indicate the presence of target gene and visual detection can be achieved.

Visual detection by fluorescence

Calcein in Loopamp Fluorescent Detection Reagent was initially combined with manganese ions to achieve the quenching effect. The amplification generates the by-product, pyrophosphate ions, which will bind and remove manganese ions from calcein to irradiate fluorescence. The fluorescence is further intensified as calcein combines with magnesium ions. From this feature, the presence of fluorescence can indicate the presence of target gene and visual detection can be achieved.

Real-time detection by turbidity

The turbidity of magnesium pyrophosphate, a by-product of the reaction, can be real-time detected by a real-time turbidimeter.
